Abstract:
Objective:To investigate the expression of circular RNA hsa_circ_0006528 (circPRELID2) with coding function in colorectal cancer (CRC) tissues and its effects on cell proliferation and apoptosis.Methods:The expression of circPRELID2 in CRC tissues was determined by in situ hybridization (ISH) and the correlation between the expression of circPRELID2 in CRC and clinicopathological parameters was analyzed.The expression of circPRELID2 in CRC tissues and cell lines were confirmed by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR).The peptide-coding potential of circPRELID2 was predicted by the circRNADb online database,and Western blot (WB) verified its presence.MTT and flow cytometry (FCM) were performed to detect how circPRELID2 and circPRELID2-encoded peptide affected the proliferation and apoptosis of CRC cells,respectively.Results:circPRELID2 was highly expressed in CRC tissues (P<0.05),and its high level was associated with a reduced survival time in CRC patients (P<0.001).circPRELID2 was highly expressed in CRC cell lines HT-29,Caco2,SW48,SW480,RKO,HCT116 and SW620 compared to normal intestinal epithelial cells HIEC (P<0.01).circPRELID2 knockdown inhibited the proliferation and promoted the apoptosis of SW620 and HCT116 cells while overexpression did the opposite (P<0.01).The WB assay confirmed the novel peptide encoded by circPRELID2 and that the peptide significantly promoted proliferation and inhibited apoptosis in SW620 and HCT116 cells (P<0.01).Conclusion:circPRELID2 with coding function has tumor promoter function in CRC and it may serve as a potential therapeutic target for CRC.
