Abstract:
Objective:To investigate the differential expression,prognostic value and biological function of treacle ribosome biogenesis factor 1(TCOF1) in hepatocellular carcinoma(HCC) by bioinformatics analysis,immunohistochemistry and clinical data analysis.Methods:To analyze the expression of TCOF1 in HCC tissues and its relationship with prognosis using data from the cancer genome atlas (TCGA).Immunohistochemistry was used to detect the expression of TCOF1 in 107 HCC patients and analyze its clinical significance.Kaplan-Meier curve analysis was applied to investigate the relationship between TCOF1 expression in HCC tissues and prognosis.We perform univariate and multivariate Cox regression analysis to identify independent factors associated with prognosis in HCC patients,and construct a predictive model,and draw Nomogram to visualization prediction models.Gene set enrichment analysis(GSEA) was used to explore the biological functions of TCOF1.Results:The expression level of TCOF1 in HCC tissues was significantly higher than that in normal liver tissues(P<0.001),and high expression was associated with poor prognosis of patients (P<0.05).Univariate and multivariate Cox regression analysis revealed that high expression of Fibrinogen(Fg) (HR=1.520,95%CI=1.166~1.981,P=0.002), PATA (HR=1.048,95%CI=1.004~1.094,P=0.031), and TCOF (HR=11.126,95%CI=1.002~1.265,P=0.046) were independent risk factors for the prognosis of HCC patients.A prognostic model was constructed and a column chart was drawn to predict the 1-,3-,and 5-year OS of HCC patients.Immunohistochemical detection confirmed positive expression of TCOF1 in HCC tissues while weakly positive or negative expression in adjacent liver tissues.GSEA enrichment analysis results suggested that TCOF1 may be involved in nine signaling pathways including oocyte meiosis and the p53 signaling pathway.Conclusion:TCOF1 is highly expressed in HCC tissues and is closely related withpoor prognosis of patients,suggesting that TCOF1 may be a potential biomarker for evaluating the prognosis of patients with HCC.
