Abstract:
Objective:To explore the possible mechanism of baicalin against proliferation,migration and invasion of uterine leiomyoma cells.Methods:Primary uterine leiomyoma cells were cultured in vitro and identified by α-Actin immunostaining.After the intervention of baicalin with 5,10 and 20 μmol/L,the survival rate of cells was detected by CCK8.The migration and invasion of cells were detected by scratch and Transwell assay.Hoechst staining was used to detect cell apoptosis.The expression of apoptosis-related proteins Bax,Bcl-2 and PI3K/AKT signaling pathway was detected by Western Blot.Results:α-Actin immunostaining identified the successful extraction of primary uterine leiomyoma cells.The results of CCK8 experiment showed that baicalin at 10 and 20 μmol/L could inhibit the growth of uterine leiomyoma cells in a dose-dependent manner.The results of scratch and Transwell experiments showed that 10 and 20 μmol/L baicalin could significantly inhibit the migration and invasion of uterine leiomyoma cells.Hoechst staining showed that 10 and 20 μmol/L baicalin significantly promoted the apoptosis of uterine leiomyoma cells.Baicalin at 20 μmol/L significantly promoted the expression of apoptotic protein Bax,and significantly inhibited the expression of anti-apoptotic protein Bcl-2.In addition,20 μmol/L baicalin significantly inhibited the expression of NF-κB,p-AKT and p-PI3K in uterine leiomyoma cells,and significantly inhibited the phosphorylation of PI3K/AKT signaling pathway.Conclusion:Baicalin significantly inhibited the proliferation,migration and invasion of uterine leiomyoma cells,and its inhibitory effect was related to the regulation of PI3K/AKT signaling pathway and phosphorylation of IκB.
