Abstract:
Objective:To investigate the expression and biological role of dipeptidyl peptidase III (DPP3) in oral squamous cell carcinoma (OSCC).Methods:By utilizing bioinformatics techniques,the differential expression of DPP3 in OSCC and its diagnostic and prognostic value were analyzed.qRT-PCR was used to detect the expression of DPP3 mRNA in OSCC tissues.By employing small interfering RNA (siRNA) technology to knock down DPP3 in CAL27,SCC15 cell lines,the knockdown efficiency was validated through qRT-PCR and Western Blot experiments.After DPP3 knockdown,CCK8,EdU,scratch,transwell migration assays,and flow cytometry were used to assess proliferation,migration capabilities,and apoptosis rates of CAL27 and SCC15 cells.Western Blot experiment to detect the expression of p65 and phosphorylated p65 protein (p-p65) in the aforementioned cell lines.Results:DPP3 expression elevated in OSCC (P<0.05).The area under the ROC curve (AUC) was 0.903 (95%CI:0.860~0.947),with sensitivity and specificity of 81.5% and 90.6%,respectively.Survival analysis revealed lower survival rates and poorer prognosis in the high-expression group compared to the low-expression group (P<0.05).After DPP3 knockdown,proliferation and migration abilities of CAL27,SCC15 cells decreased,while apoptosis rates increased (P<0.05).The p65/GAPDH ratio showed no significant change (P>0.05),while the p-p65/p65 ratio decreased (P<0.05).Conclusion:DPP3 is upregulated in OSCC tissues,holding diagnostic and prognostic significance,promoting proliferation and migration capabilities while inhibiting apoptosis in OSCC cells,potentially functioning through the NF-κB signaling pathway.
