Abstract:
Objective:To investigate the mechanism of reversal of cisplatin resistance in gastric cancer by Zuojin Wan.Methods:In vitro,use human gastric cancer cell line SGC-7901 and human gastric cancer cisplatin-resistant cell line SGC-7901/DDP.The cell proliferation rate was detected by CCK-8 assay.The protein expression of each group were detected by Western blot and quantitative real-time fluorescence PCR(qRT-PCR) after the intervention of Zuojin Wan alcohol extract.Meanwhile,RTKN gene lentiviral vector-transfected effect was examined by combining CCK-8,qRT-PCR and Western blot assays.In vivo,we transplanting RTKN-interfered SGC-7901/DDP cells into nude mice subcutaneously and giving Zuojin Wan alcohol extract intervention for 4 weeks respectively.The size of transplanted tumours was recorded and tumour growth curves were plotted,and the expression of RTKN,p53,Ace-p53 and HDAC1 proteins were detected by Western blot.Results:RTKN expression was significantly higher in SGC-7901/DDP cells than in SGC-7901,and silencing RTKN significantly reduced drug resistance in SGC-7901/DDP cells.Zuojin Wan significantly reduced drug resistance in SGC-7901/DDP cells,but after silencing of RTKN had no significant effect on the resistance of SGC-7901/DDP cells and on the volume of subcutaneous transplanted tumors in nude mice,suggesting that the antitumor effect of Zuojin Wan was suppressed after reducing RTKN expression.Conclusion:RTKN gene can increase the tolerance of SGC-7901/DDP cells to cisplatin.Zuojin Wan can effectively increase the inhibitory effect of chemotherapeutic drugs on the proliferation of gastric cancer cells,reduce the expression of RTKN protein in gastric cancer resistant cells,and reverse the cisplatin resistance of gastric cancer.
