Abstract:
Objective:To investigate the expression of WD40-encoding RNA antisense to p53(WRAP53)also known as telomerase cajal body protein 1(TCAB1) in papillary thyroid carcinoma(PTC) and its relationship with clinical pathological characteristics and prognosis.Methods:Fresh tissue samples of PTC and adjacent normal tissues were collected from 28 patients,and quantitative real-time polymerase chain reaction(qPR-PCR) and immunohistochemistry were used to detect the expression of WRAP53 mRNA and protein in PTC and surrounding normal tissues.Clinical data and corresponding paraffin sections of cancer tissues were collected from 117 PTC patients after surgery for immunohistochemistry detection,and the correlation between the protein expression level and clinical pathological characteristics of PTC was analyzed.Finally,The Human Protein Atlas(HPA) database was used to explore the relationship between WRAP53 and the prognosis of PTC.Results:The results of qPR-PCR and immunohistochemistry showed that WRAP53 was overexpressed in papillary thyroid carcinoma(PTC) compared to normal tissues(P<0.05).Compared with PTC patients with low expression of WRAP53,PTC patients with high expression of WRAP53 had larger tumor size and were more likely to have lymph node metastasis(P<0.05).The HPA database showed that the survival rate of thyroid cancer patients with high expression of WRAP53 was lower than that of patients with low expression of WRAP53,and the difference was statistically significant(P=0.041).Conclusion:This study demonstrates that WRAP53 is overexpressed in PTC tissues compared to surrounding normal tissues,and its high expression is associated with proliferation,lymph node metastasis,and poor prognosis of PTC.WRAP53 may be a new potential therapeutic target for the treatment of PTC.
