Abstract:
Objective:To investigate the impact of CircMATR3 on the malignant biological behavior of nasopharyngeal carcinoma cells by regulating miR-129-5p/SOX2 axis.Methods:Nasopharyngeal carcinoma CNE1 cells were separated into NC group,si-NC group,si-CircMATR3 group,miR-NC group,miR-129-5p mimic group,si-CircMATR3+inhibitor NC group,si-CircMATR3+miR-129-5p inhibitor group,and si-SOX2 group.RT-qPCR was applied to detect the expression of CircMATR3,miR-129-5p and SOX2 mRNA in cells.Western blot was applied to detect the protein expression.The targeting relationship between CircMATR3 and miR-129-5p,miR-129-5p and SOX2 was verified by double luciferase report experiment.Clonogenic assay was applied to detect the ability of cell cloning.Hoechst33342 assay was used to observe the morphological changes of cells.Transwell chamber test was applied to detect the cell invasion ability.Cell cycle and apoptosis was detected by flow cytometry.Results:There was a targeting relationship between CircMATR3 and miR-129-5p,between miR-129-5p and SOX2.Silencing CircMATR3 or overexpressing miR-129-5p or silencing SOX2 was able to inhibit the cloning and invasion and cell cycle transition of nasopharyngeal carcinoma cell CNE1,and promote cell apoptosis and apoptotic body formation.Inhibition of miR-129-5p expression could reverse the inhibition of silencing CircMATR3 on CNE1 cells to some extent.Conclusion:Silencing CircMATR3 can up regulate the expression of miR-129-5p and inhibit the expression of SOX2,thereby inhibiting the cloning and invasion of nasopharyngeal carcinoma cell CNE1 and promoting cell apoptosis.
