Abstract:
Objective:To explore the expression and biological functions of MTM1 (myotubularin 1) in the promotion of liver cancer metastasis.Methods:Bioinformatic analysis and immunohistochemistry staining assay were applied to determine the expression and prognostic significance of MTM1 in liver cancer.The effect of forced MTM1 expression on migration of liver cancer cells was determined by wound healing and Transwell migration assays.The effect of forced MTM1 expression on invasion of liver cancer cells was determined by Transwell invasion assay.The effect of forced MTM1 expression on the expressions of epithelial-mesenchymal transition(EMT) markers was determined by real-time-PCR and Western blot analysis.The effect of forced MTM1 expression on the in vivo metastasis of liver cancer cells was determined by nude mice lung metastasis model.Results:Bioinformatic analysis and immunohistochemistry staining assay revealed that MTM1 expression was markedly decreased in tumor tissues of liver compared non-tumor tissues,especially in metastatic tumor tissues.In addition,patients with low MTM1 expression had significant poorer survival than patients with high MTM1 expression.The migration ability of liver cancer cells was significantly suppressed when MTM1 was overexpressed.The invasion ability of liver cancer cells was significantly decreased when MTM1 was overexpressed.The progress of EMT was inhibited by MTM1 overexpression,as evidenced by increased expressions of epithelial marker (E-cadherin and ZO-1) and decreased expressions of mesenchymal marker (N-cadherin and Vimentin).Forced MTM1 expression significantly impaired the in vivo metastasis of liver cancer cells.Conclusion:MTM1 downregulation promotes the progression of EMT and thus migration and invasion in liver cancer cells.
