Abstract:
Objective:To investigate the expression of PHF1 in patients with colorectal cancer (CRC) and its effects on the proliferation,apoptosis,invasion and epithelial mesenchymal transformation (EMT) of colorectal cancer cells.Methods:SW480 and HCT 116 cells were divided into negative control group (si-PHF1-NC group,transfected with si-PHF1-NC) and experimental group (si-PHF1 group,transfected with si-PHF1).Real-time fluorescent quantitative PCR (RT-qPCR) was used to detect the level of PHF1 mRNA in CRC tissues and cells.The expression of PHF1 protein in CRC tissues was detected by immunohistochemistry.The proliferation activity of CRC cells was detected by MTT assay.The number of CRC cell clones was detected by clonogenic assay.The apoptosis of CRC cells was detected by flow cytometry.The migration and invasion of CRC cells were detected by Transwell cell assay.Western blot was used to detect the expression of E-cadherin,Vimentin and N-cadherin in CRC cells.Results:Compared with the adjacent tissues,the PHF1 mRNA level and the number of positive cells in CRC tissues increased obviously (P<0.05).Compared with NCM460 cells,the level of PHF1 mRNA in SW480 and HCT 116 cells of CRC increased obviously (P<0.05).Compared with the si-PHF1-NC group,the survival rate,numbers of migration and invasion,the protein expressions of Vimentin,N-cadherin of SW480 and HCT 116 cells in the si-PHF1 group decreased obviously (P<0.05),the poptosis rate and the protein expression of E-cadherin increased obviously (P<0.05).Conclusion:PHF1 is highly expressed in CRC tissues and cells,it can promote the proliferation and EMT of CRC cells,and inhibit apoptosis.
