Abstract:
Objective:To investigate the effects of Plexin-B1 interference on autophagy and cisplatin sensitivity of ovarian cancer SKOV3 cells.Methods:Plexin-B1 siRNA sequence and NC sequence were synthesized and transfected into ovarian cancer SKOV3 cells,and the transfection efficiency was detected by qRT-PCR.The cells were divided into control group,NC group and si-Plexin-B1 group.The cell activity was detected by CCK8,and the expression of Plexin-B1 mRNA was detected by qRT-PCR.The protein expressions of Plexin-B1,Akt,p-Akt,LC3-Ⅱ,CALR and PRKDC were detected by Western blot.Cells were treated with different concentrations of cisplatin(1,2,4,8,16,32 μg/mL),the proliferation of cells in each group was detected by CCK8,and the effect of interfering Plexin-B1 on cisplatin sensitivity was evaluated.Results:Compared with the control group,the activity of ovarian cancer SKOV3 cells,the expression levels of Plexin-B1 mRNA and protein,and the phosphorylation level of Akt protein were significantly decreased after Plexin-B1 interference(P<0.05),the protein expression levels of LC3-Ⅱ,CALR and PRKDC were significantly increased(P<0.05).CCK8 results showed that the proliferation ability of ovarian cancer SKOV3 cells gradually decreased with the increase of cisplatin concentration,and the proliferation ability of ovarian cancer SKOV3 cells after Plexin-B1 interference was lower than that of the control group,that is,the sensitivity of ovarian cancer SKOV3 cells to cisplatin increased after Plexin-B1 interference.Conclusion:Plexin-B1 can promote the autophagy of ovarian cancer SKOV3 cells through PI3K/Akt pathway,and thus improve the sensitivity of ovarian cancer SKOV3 cells to cisplatin.
