Abstract:
Objective:To investigate the expression of KIF14 in intrahepatic cholangiocarcinoma (ICC) and its effects on the biological behavior of cholangiocarcinoma cells.Methods:The mRNA expression level of KIF14 in ICC tissue and paracancerous tissue was detected by qRT-PCR.The protein expression of KIF14 in ICC tissue and paracancerous tissue was detected by immunohistochemistry,and the relationship between KIF14 expression and clinical pathological characteristics was analyzed.The siRNA technology was used to inhibit the expression of KIF14 in QBC939 cells and the inhibitory effect was detected.Western blotting assay was used to detect the effects of inhibiting the expression of KIF14 on AKT signaling pathway.We performed MTT and Transwell assay to detect the ability of proliferation and invasion after inhibiting the expression of KIF14 in QBC939 cells.Results:The result of qRT-PCR indicated that the mRNA expression level of KIF14 in ICC tissue was significantly higher than that in the paracancerous tissue.The result of immunohistochemistry staining indicated that the protein expression level of KIF14 in ICC tissue was higher than that in the paracancerous tissue.The expression level of KIF14 was related to the tumor size,lymph node metastasis and TNM stage.The siRNA technology could effectively inhibit the mRNA expression of KIF14 in QBC939 cells and decrease the phosphorylation level of AKT.The results of MTT assay and Transwell assay showed that inhibiting the expression of KIF14 can significantly reduce the ability of proliferation and invasion of QBC939 cells.Conclusion:KIF14 is highly expressed in ICC tissue and its high expression is associated with adverse clinical and pathological characteristics.KIF14 promotes the proliferation and invasion of cholangiocarcinoma cells through AKT signaling pathway.
