Abstract:
Objective:To investigate the effects of Cisplatin (CDDP) combined with Rosmarinic acid (RA) on the proliferation and apoptosis of human primary hepatocellular carcinoma Huh7 hepatoma cells.Methods:The IC50 values after 24 h,48 h and 72 h treatment with CDDP and RA respectively were detected and calculated by CCK-8 cell proliferation assay.Then,drug untreated group (control group),5 μg/mL CDDP group,200 μg/mL RA group,and 5 μg/mL CDDP combined with 200 μg/mL RA group were set.The apoptosis rates of the control group,CDDP group,RA group and combination group were (7.28±0.56)%,(18.23±2.53)%,(26.44±1.99)% and (37.27±0.28)% by flow cytometry,respectively.Finally,the expression levels of Bcl-2,Bax,Cleaved-caspase-3 and Procaspase-3 apoptosis-related proteins were detected by Western blot.Results:RA enhanced the inhibitory ability of CDDP on the proliferation of Huh7 cells and showed a concentration-dependent relationship (P<0.05),and the apoptosis rate of Huh7 cells was significantly increased in the CDDP combined with RA-treated group (P<0.05),and the expression levels of the pro-apoptotic proteins Bax and Cleaved-caspase-3 increased after the effect of CDDP combined with RA,while expression levels of Bcl-2 were decreased (P<0.05).Conclusion:RA can inhibit the proliferation of Huh7 cells and induce cell apoptosis,and has a synergistic effect with Cisplatin.The mechanism may be related to the activation of mitochondrial apoptosis pathway.
