Abstract:
Objective:To study the effects of myeloma overexpressed gene (MYEOV) on the proliferation,migration,and invasion of pancreatic cancer cells,as well as the regulation of epithelial-mesenchymal transformation (EMT) related proteins and STAT3 signal pathway-related proteins.Methods:The UCSC Xena database was used to analyze the expression level of MYEOV in pancreatic cancer and normal tissues,and the ICGC database was used to analyze the relationship between MYEOV and the prognosis of pancreatic cancer patients.A lentivirus recombinant PaTu8988 cell line with stable low expression of MYEOV was constructed.Quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot were used to detect MYEOV mRNA and protein expression in the pancreatic cancer cell line.CCK-8,scratch assay,Transwell migration,and invasion assay were used to analyze cell proliferation,migration,and invasion ability.The expressions of E-cadherin,Vimentin,MMP2,MMP9,STAT3,and p-STAT3 were detected by Western blot.Results:Bioinformatics analysis showed that MYEOV was highly expressed in pancreatic cancer tissues and was associated with a poor prognosis of pancreatic cancer.Compared with the control group,the proliferation,migration,and invasion ability of pancreatic cancer cell line PaTu8988 decreased after the down-regulation of MYEOV.Compared with the control group,after the down-regulation of MYEOV,E-cadherin protein expression was increased in the PaTu8988 cell line,and Vimentin,MMP2,MMP9,and p-STAT3 protein expression was down-regulated.Conclusion:The down-regulation of MYEOV expression inhibits the proliferation,migration,and invasion of pancreatic cancer cells,and the mechanism may be related to the inhibition of EMT.The change of STAT3 pathway protein during EMT suggests that the STAT3 pathway may be involved in regulating this phenotype.
