Abstract:
Objective:To analyze the effect of targeted regulation of VEGFR-2/STAT3 on malignant obstructive jaundice(MOJ) rats.Methods:8-week SPF SD rats were randomly grouped into 5 groups:Sham group(group A),MOJ group(group B),and apatinib low-dose group(42.5 mg/kg,group C),apatinib medium-dose group(85.0 mg/kg,group D),apatinib high-dose group(170.0 mg/kg,group E),10 rats in per group.The group A:The rats were underwent sham operation.The group B,C,D and E:The tumor mass cultured from Walker-256 liver cancer strain was implanted into the junction of the middle hepatic lobe and the right hepatic lobe at a distance of 5 mm from the hepatic hilum in order to cause cancerous stricture of the bile duct to prepare MOJ.The group C,D and E:The MOJ rats were given apatinib solution by gavage every day.The group A and B:The rats were given normal saline.After 5 weeks of treatment,the general condition of the rats was observed,the liver function indexes and serum liver enzyme indexes were detected,and the Western blot was used to detect the p-VEGFR-2,VEGFR-2,p-STAT3,STAT3,Bax and Bcl-2 in the transplanted tumors.Results:Compared with the group A,the total bilirubin(TBIL),direct bilirubin(DBIL),indirect bilirubin(IBIL),alkaline phosphatase(ALP),aspartate aminotransferase(AST) and alanine aminotransferase(ALT) in group B had increased(P<0.05).Compared with the group B,the TBIL,DBIL,IBIL,AST,ALP,ALT and tumor growth rate had significantly decreased in the group C,D,E,and the group E had the most significant decrease(P<0.05).Compared with the rats in the group A,the expression of p-VEGFR-2,p-STAT3 and Bax in group B had significantly increased,and the expression of Bcl-2 had significantly decreased(P<0.05).Compared with the rats in the group B,the expression of VEGFR-2,p-STAT3,Bax had significantly decreased,and the expression of Bcl-2 was significantly increased in the group C,D,E,and the decreased/increased level were the most significant in the group E(P<0.05).There was no statistical difference in VEGFR-2 and STAT3 in 5 groups(P>0.05).Conclusion:The possible mechanism of apatinib in the treatment of MOJ is to promote the apoptosis of cancer cells,which is related to the targeted regulation of the VEGFR-2/STAT3,thereby effectively alleviating the obstruction of hepatic microbiliary ducts in MOJ,and reducing hepatocyte and liver injury.The targeted regulation of apatinib in the treatment of MOJ is in a dose-dependent manner,apatinib at 170.0 mg/kg was the most effective.
