Abstract:
Objective:To investigate the effect of different methods of preservation of detection of BRAF V600E mutation in fine needle aspiration cytology(FNAC) of thyroid.Methods:Retrospective analyzed study of 164 patients from Jan 2019 to Feb 2022 who underwent thyroid fine needle aspiration through ultrasound-guided in our hospital.DNA extracted respectively from materials preserved in commercial cell preservation solution,normal saline preservation solution and stained smears is used for the detection of BRAF V600E mutation.Results:There are no significant differences in the mean DNA concentration and the Ct value of reference gene among the three methods(P>0.05).For the A260/280 value,there was no statistically significant difference between the cell preservation solution group and the saline preservation solution group,nor between the saline preservation solution group and the scraping slice group (P>0.05),while there was statistically significant difference in the A260/280 value between the cell preservation solution group and the scraping slice group(P<0.05).Conclusion:Scraping thyroid FNAC samples can obtain high DNA purity and low laboratory requirements,which is suitable for promotion in primary hospitals.
