Abstract:
Objective:To investigate the effects of reticulin 4B (Nogo-B) and M2 polarization of macrophages on proliferation,invasion and angiogenesis in ovarian cancer and its related mechanisms.Methods:26 cases of ovarian cancer tumor tissues and adjacent in our hospital were collected,and the protein expression of Nogo-B detected by immunohistochemistry.Macrophages from tumor and adjacent tissues were isolated,and the proportion of CD86 and CD206 phenotype and Nogo-B expression of macrophages were detected by flow cytometry and qRT-PCR.si-Nogo-B and si-NC were transfected into A2780 cells by liposome transfection method.The expression of Nogo-B and VEGF in A2780 cells detected by qRT-PCR and Western blot,while the proliferation activity detected by CCK8 assay.THP-1 cells were divided into 3 groups and examined their Nogo-B expression levels:si-NC-PMA group,si-NC-PMA/IL4 group,si-Nogo-B-PMA/IL4 group (after transfection of si-Nogo-B and si-NC into THP-1 cells,then cell polarization was induced by PMA or PMA/IL4).Macrophages in each group were co-incubated with A2780 cells.Proliferation,invasion and VEGF expression of A2780 cells was detected by CCK8,Transwell,qRT-PCR and Western blot respectively.Results:Nogo-B expression was increased at tumor sites in patients with ovarian cancer compared with adjacent tissues.Macrophages at the tumor site showed M2 type polarization,and it's Nogo-B expression was significantly increased.There was no significant difference in VEGF expression level and proliferation ability in A2780 cells between si-Nogo-B group and si-NC group.Compared with si-NC-PMA group,macrophages in si-NC-PMA/IL4 group showed M2-type polarization,and the expression of Nogo-B was increased,and the proliferation and invasion ability of A2780 cells co-incubated with si-NC-PMA/IL4 group were increased,as well as the expression level of VEGF.Compared with si-NC-PMA/IL4 group,the proportion of M2-type macrophages and the expression of Nogo-B in si-Nogo-B-PMA/IL4 group were decreased,and the proliferation and invasion ability of A2780 cells co-incubated with si-Nogo-B-PMA/IL4 group were weakened,as well as the expression level of VEGF.Conclusion:Nogo-B may induce proliferation,invasion and angiogenesis of ovarian cancer by promoting M2-type polarization in macrophages.
