Abstract:
Objective:To study the expression levels of hematopoietic stem cell specific associated binding protein-1(HAX1) and mutant-p53 expression in endometrial carcinoma(EC),and the correlation with prognosis of EC.Methods:UALCAN database and qRT-PCR were used to check the expression levels of HAX1 mRNA in fresh EC tissues.Western blotting was used to check the expression levels of HAX1 protein in fresh EC tissues.Immunohistochemical(IHC) method was used to detect the proteins levels of HAX1 and p53 in a total of 120 paraffin EC and adjacent normal tissues and their correlation.The relationships between HAX1 and p53 and clinicopathological features of EC and the prognosis of patients were further analyzed.GEPIA database was used to further analyze the correlation between HAX1 and EC related genes.Results:HAX1 mRNA and protain were highly expressed in EC.The protein expressions of HAX1 and mutant-p53 were significantly positively correlated(χ2=36.468,r=0.551,P<0.05),and both were closely related to lymph node metastasis,FIGO staging,blood CEA level and Ki-67 expression(P<0.05),but negatively correlated with overall survival of patients with EC.HAX1 positive expression(P=0.008),p53 mutant expression(P=0.022),lymph node metastasis(P=0.003),FIGO high stage(P<0.001)and postoperative adjuvant therapy(P<0.001) were independent risk factors for prognosis of EC patients.Also,HAX1 was correlated with the expression of TP53,MKI67,BAX,CCNE1,POLE1,MSH2,MSN6 and PMS2.Conclusion:HAX1 is highly expressed in EC,and positively correlated with mutant-p53 expression.The expression mode of the two could comprehensively evaluate the malignancy,progression of the cancer and prognosis of the patients,providing new ideas for the postoperative treatment of EC.
