Abstract:
Objective:To investigate the effects of miR-515-5p on the proliferation,migration and cisplatin resistance of laryngeal carcinoma cells and to predict its potential target genes.Methods:In laryngeal cancer cells FD-LSC-1 and AMC-HN-8,the expression of miR-515-5p was detected by reverse transcription quantitative PCR.After knocking down and over-expressing miR-515-5p,cell proliferation was detected by cell counting kit 8 and clone formation assay,cell migration was detected by Transwell assay,and the sensitivity of cells to cisplatin was detected by CCK8 method.The model of transplanted tumor in nude mice was established,and the effect of knocking down miR-515-5p on the growth of transplanted tumor was detected by in vivo imaging,tumor weighing and immunohistochemical experiment.The potential target genes of miR-515-5p were predicted by four miRNA target gene databases,laryngeal squamous cell carcinoma microarray datasets and RT-qPCR experiment.Results:The expression of miR-515-5p was significantly increased in FD-LSC-1 and AMC-HN-8 cells(P<0.001).After knocking down miR-515-5p,the proliferation ability,clone formation number,cell migration number and cell viability after cisplatin treatment of FD-LSC-1 and AMC-HN-8 cells were significantly decreased(P<0.05).The photon intensity and tumor weight of transplanted tumor in nude mice were also significantly decreased(P<0.05),and the expression of proliferation marker Ki-67 was decreased.Overexpression of miR-515-5p could significantly promote the proliferation,migration and cisplatin resistance of laryngeal cancer cells(P<0.05).There was a negative regulatory relationship between miR-515-5p and poly(A) binding protein interacting protein 2B(P<0.05).Conclusion:miR-515-5p may promote the proliferation,migration and cisplatin resistance of laryngeal carcinoma cells by regulating the target gene PAIP2B,and play a role in targeted therapy combined with cisplatin chemotherapy of laryngeal carcinoma.
