Abstract:
Objective:To investigate the effect and mechanism of miR-1471 on proliferation,invasion and migration of glioma cells.Methods:Human glioma cells U251 were cultures in vitro.miR-1471 mimics or negative control was transfected into U251 cells,respectively,and were recorded as miR-1471 group and negative control(NC) group,respectively.Control group was set.qRT-PCR was used to detect the transfection effect.CCK8 was used to detect cell proliferation.Transwell assay was used to detect the invasive ability of cells in each group.The scratch test was used to detect the migration ability of cells in each group.qRT-PCR and Western blot were used to detect MTDH and Wnt/β-catenin signaling pathway related genes and proteins.Results:Compared with control group and NC group,the proliferation activity was significantly decreased(P<0.05),the number of invasive cells was significantly decreased(P<0.05),the scratch spacing increased significantly,mRNA and protein expression levels of MTDH,Wnt1 and β-catenin was significantly decreased(P<0.05) in miR-1471 group.Conclusion:miR-1471 can inhibit the proliferation,invasion and migration of human glioma cells U251,and its mechanism may be related to down-regulation of MTDH expression and inhibition of activation of Wnt/β-catenin signaling pathway.
