Abstract:
Objective:To investigate the effect of downregulation of receptor for advanced glycation end products (RAGE) on the proliferation,apoptotic capacity and cycle distribution of bladder cancer T24 cells.Methods:After the human bladder cancer T24 cell line was cultured,the down-regulated RAGE group and the up-regulated RAGE group were constructed by lentiviral vector.Cell proliferation ability was detected by MTT,cell apoptosis and cycle distribution were detected by flow cytometry,cell invasion ability was detected by Transwell chamber test,cell migration ability was detected by cell scratch test,and relative expression of CDK4,p-Akt,ICAM-1,MMP-9,Bcl-2,Bak and caspase7 was detected by Western blot.Results:At 24,48 and 72 h,the cell proliferation rate in the up-regulated RAGE group was higher than that in the control group and the down-regulated RAGE group,and the apoptosis rate was lower than that in the control group and the down-regulated RAGE group (P<0.05).The proportion of cells in the up-regulated RAGE group in the G0/G1 phase was lower than that in the control group and the down-regulated RAGE group (P<0.05).The relative expression of CDK4,p-Akt,ICAM-1,MMP-9,Bcl-2,caspase7 protein and the number of invasive cells and migrating cells in RAGE group were lower than those in control group,and the relative expression of Bak protein was higher than that in control group.The relative expression of CDK4,p-Akt,ICAM-1,MMP-9,Bcl-2,caspase7 protein and the number of invasive cells and migrating cells in RAGE group were higher than those in control group,and Bak protein was lower than that in RAGE group.The relative protein expression was lower than that in the control group (P<0.05).Conclusion:Downregulation of RAGE expression can inhibit the proliferation of bladder cancer T24 cells,regulate the relative expression of p-Akt,CDK4,ICAM-1,MMP-9,block the cell cycle distribution,inhibit cell invasion and migration,regulate the relative expression of caspase7,Bcl-2,Bak,and promote the apoptosis of bladder cancer T24 cells.
