Abstract:
Objective:To investigate the effect of miR-106b-5p on the proliferation,migration and invasion of colorectal cancer(CRC) cells and its underlying mechanism.Methods:The expression levels of miR-106b-5p in CRC tissues and adjacent tissues,immortalized intestinal epithelial cells and CRC cells were detected by qPCR.CCK8 assay,scratch assay and Transwell assay were used to detect the proliferation,migration and invasion ability of DLD1 cells.The downstream target gene of miR-106b-5p was searched by bioinformatics method.Dual luciferase assay and RNA binding protein immunoprecipitation assay was conducted to determine the relationship between miR-106b-5p and cyclin D1(CCND1).The level of CCND1 protein in cells and tissues was detected by Western blotting assay.Results:Compared with adjacent tissues,the expression level of miR-106b-5p in colorectal cancer tissues was decreased(P<0.05).The expression level of miR-106b-5p in colorectal cancer cells was decreased compared with that in immortalized intestinal epithelial cells(P<0.05).CCK8 assay,scratch assay and Transwell assay showed that overexpression of miR-106b-5p significantly inhibited the proliferation,migration and invasion in DLD1 cells(P<0.05).Starbase database predicts that the downstream target gene of miR-106b-5p was CCND1,and miR-106b-5p acted on the 3'-UTR region of CCND1.Dual luciferase assay and RNA binding protein immunoprecipitation assay confirmed the relationship between miR-106b-5p and CCND1.CCND1 was highly expressed in colorectal cancer tissues and cells(P<0.05),and the overexpression of miR-106b-5p significantly inhibited CCND1 expression in DLD1 cells(P<0.05).Conclusion:Decreased level of miR-106b-5p can inhibits proliferation,migration and invasion of colorectal cancer cells via targeting CCND1.
