Abstract:
Objective:To explore the expression and significance of USP18 in cervical squamous cell carcinoma,and to analyze the relationship between the expression level of USP18 in cervical cancer and its clinical application value.Methods:The mRNA expressions of USP18 in 30 cases of fresh cervical cancer tissues and the paracancerous normal cervical tissues were detected by real-time fluorescence quantitative PCR, and the protein expressions of USP18 in 6 cases of fresh cervical cancer tissues and the paracancerous normal cervical tissues were detected by western blotting,while the protein expression of USP18 in 60 paraffin-embedded cervical cancer specimens and paracancerous normal cervical specimens was detected by immunohistochemistry.The relationship between the expression of USP18 and the clinical pathological characteristics of cervical cancer was analyzed.Results:In fresh tissues,the mRNA and protein levels of USP18 in cervical cancer tissues were significantly higher than those in normal cervical tissues(P<0.01,P<0.01).In paraffin-embedded specimens,the expression of USP18 in cervical cancer tissues was significantly higher than it in normal cervical tissues(86.7% vs 33.9%,P<0.01),and the relative expression of USP18 was closely associated with differentiation and lymph node metastasis(P<0.01),but no statistical difference was observed between the relative expression of USP18 and age(P>0.05).Conclusion:The expression of USP18 is significantly increased in cervical cancer and it is related to the differentiation and metastasis of cervical cancer,which is expected to be a new molecular marker of cervical cancer.
