Abstract:
Objective:To explore the expression and clinical significance of long non-coding RNA nucleolar small molecule RNA host gene 1(SNHG1) in cholangiocarcinoma.Methods:RT-PCR was used to detect the relative expression levels of SNHG1 in 64 cases of cholangiocarcinoma tissues and their paired adjacent tissues,cholangiocarcinoma cells(RBE and QBC939) and normal intrahepaticbile duct epithelial cells(HIBE).The relationship between SNHG1 and the clinicopathological characteristics of cholangiocarcinoma was analyzed,and the relationship between SNHG1 and the survival and prognosis of patients with cholangiocarcinoma was analyzed by COX multivariate analysis and Kaplan-Meier survival curve.SNHG1 overexpression plasmid(overexpression group),SNHG1 knockdown plasmid(silence group) and negative control plasmid(control group) were transferred into RBE cells with LipofectAMINETM 2000.Cell proliferation was detected by CCK8 method.Cell cycle was detected by flow cytometry.Cell invasion was detected by Transwell.Results:The relative expression of SNHG1 in cancer tissues was significantly higher than that in adjacent tissues(P<0.05).Compared with HIBE cells,the relative expression of SNHG1 in RBE and QBC939 cells washigher(P<0.05).SNHG1 was related to TNM staging and lymph node metastasis(P<0.05),but not related to gender,age,tumor location and degree of differentiation(P>0.05).COX multivariate analysis showed that TNM staging,lymph node metastasis and high expression of SNHG1 were independent factors influencing the poor survival and prognosis of patients with cholangiocarcinoma(P<0.05).The Kaplan-Meier survival curve showed that the median survival time of high expression group(17 months) was lower than that of low expression group(42 months),and the difference was statistically significant(χ2=6.698,P=0.003).Compared with control group,the relative expression of SNHG1,cell proliferation and invasion activity insilent group were significantly reduced(P<0.05),cell cycle was arrested in G1 phase.While the above indicators in overexpression group were significantly increased(P<0.05),cell cycle more enteredin G2 phase.Conclusion:SNHG1 may be involved in the occurrence and development of cholangiocarcinoma,and is related to survival prognosis.
