Abstract:
Objective:To investigate the expression and clinical significance of microRNA-206 (miR-206) and family with sequence similarity 83 member A (FAM83A) in bladder urothelial carcinoma (BUC) tissues.Methods:From February 2015 to February 2017,86 patients with BUC who underwent surgical treatment in the surgical oncology department of our hospital were selected as the research objects,and the cancer tissues and adjacent tissues were selected as BUC group and control group respectively.Real-time fluorescence quantitative PCR (qRT-PCR) was used to detect the relative expression of miR-206 and FAM83A mRNA.Immunohistochemistry was used to detect the expression of FAM83A in cancer tissues and adjacent tissues.The relationship between the expression of miR-206 and FAM83A and clinicopathological characteristics was analyzed.The relationship between the expression of miR-206 and FAM83A and the 3-year survival of patients with BUC was analyzed by Kaplan-Meier method.Results:The expression level of miR-206 in BUC group was significantly lower than that in control group (P<0.05),and the expression level of FAM83A mRNA in BUC group was significantly higher than that in control group (P<0.05).Immunohistochemical result showed that the positive rate of FAM83A in BUC group was higher than that in control group (P<0.001).The expression levels of miR-206 and FAM83A in BUC tissues were closely correlated with tumor stage,lymph node metastasis,histological grade and tumor diameter (P<0.05),but not with age and gender (P>0.05).Kaplan-Meier analysis showed that the 3-year cumulative survival rate of patients with BUC was 52.33%(45/86).The 3-year cumulative survival rate of patients with high expression of miR-206 was higher than that of patients with low expression of miR-206.The 3-year cumulative survival rate of patients with high expression of FAM83A was lower than that of patients with low expression of FAM83A.Multivariate COX analysis showed that low expression of miR-206 and high level of FAM83A were independent risk factors for poor prognosis of BUC patients (HR=2.598,3.342,P<0.05).Conclusion:The expression of miR-206 is low and the expression of FAM83A is high in the cancer tissues of BUC patients.miR-206 expression and FAM83A expression are related to tumor stage,lymph node metastasis,histological grade,tumor diameter and adverse prognosis in BUC patients.
