Abstract:
Objective:To investigate aberrant methylation of LAMA2 genes in invasive breast cancer and its adjacent bresast tissues,and analyze the correlation between clinical and pathological features.Methods:The samples were collected from 101 invasive breast cancer patients who underwent eclecive radical resection.The methylation status of LAMA2 genes in breast cancer and its adjacent tissues was quantitatively detected by pyrophosphate sequencer.The expression of LAMA2 gene was detected by RNA reverse transcription and PCR,and the relationship between LAMA2 gene and the clinical and pathological characteristics was analyzed,as well as the prognostic factors in breast cancer patients.Results:Methylation rate of LAMA2 gene in 101 cases of breast cancer was 96%.The methylation rate was higher than that of the corresponding adjacent tissues,and the difference was statistically significant(P<0.05).Meanwhile,the mRNA expression of LAMA2 was lower in breast cancer tissues with LAMA2 promoter region hypermethylated(P<0.05).The methylation of LAMA2 gene was significantly associated with tumor diameter,axillary lymph node metastasis,histological grade,HER-2(all P<0.05).Cox proportional hazard model analysis indicated that lymph node metastasis and LAMA2 gene methylation were independent prognostic factors for breast cancer(RR=32.127,1.222,95%CI=1.296~796.112,1.103~1.355,P=0.034,0.000).Conclusion:LAMA2 gene methylation and its aberrant expression might be associated with breast cancer.LAMA2 gene is closely related to the occurrence and development of breast cancer,and this is the first report of its methylation degree as a potential biological marker for malignancy and prognosis of breast cancer.
