Abstract:
Objective:To study the effects of lncRNA NR2F2-AS1 on proliferation and apoptosis of gastric cancer cells and to explore its mechanism.Methods:qRT-PCR was used to detect the mRNA expression of NR2F2-AS1 and miR-425-5p in the cells.pcDNA group（transfected pcDNA),pcDNA-NR2F2-AS1 group（transfected pcDNA-NR2F2-AS1),sh-NR2F2-AS1 group(transfected sh-NR2F2-AS1),sh-NC group(transfected sh-NC),anti-miR-NC group（transfected anti-miR-NC),anti-miR-425-5p group（transfected anti-miR-425-5p),pcDNA-NR2F2-AS1+miR-NC group（co-transfected pcDNA-NR2F2-AS1 and miR-NC),pcDNA-NR2F2-AS1+miR-425-5p（co-transfected pcDNA-NR2F2-AS1 and miR-425-5p mimics) were transfected into MGC-803,MKN-45 cells.Proliferation was detected by MTT assay.Flow cytometry detected cell apoptosis.Dual luciferase reporter assay detected fluorescence activity of cells.Results:Compared with GES-1,a normal gastric mucosal epithelial cell,the expression of NR2F2-AS1 was significantly decreased in gastric cancer cells MGC-803,MKN-45,and the expression of miR-425-5p was significantly increased.Overexpression of NR2F2-AS1 and inhibition of miR-425-5p can inhibit the proliferation of gastric cancer cells and promote apoptosis.miR-425-5p can inhibit the fluorescence activity of wild-type NR2F2-AS1 MGC-803,MKN-45 cells.Overexpression of miR-425-5p can reverse the overexpression of NR2F2-AS1 to MGC-803,MKN-45 cell proliferation and apoptosis.Conclusion:Long-chain non-coding RNA NR2F2-AS1 can inhibit the proliferation of gastric cancer cells and promote apoptosis.The mechanism may be related to targeting miR-425-5p,which will provide a new target for the treatment of gastric cancer.
