Abstract:
Objective:To detect the expression of miRNA-194-5p(miR-194-5p) and E-cadherin in cholangiocarcinoma,explore the relationship between miR-194-5p and E-cadherin.Methods:49 cases of cholangiocarcinoma were selected as the observation group,and 49 cases of normal bile duct epithelial tissue were selected as the control group.Expression of miR-194-5p was detected by real-time fluorescent quantitative PCR(qPCR) in the two groups.Expression of E-cadherin and N-cadherin were detected by immunohistochemistry in the observation group.We established blank control group,empty vector group and miR-194-5p transfection group of cholangiocarcinoma cell line.Expression of E-cadherin and N-cadherin were detected by Western Blot methods.Results:Expression of miR-194-5p was significantly lower in observation group than that in the control group.Expression of miR-194-5p was statistically significant in maximum diameter,nerve involvement,differentiation degree,metastasis and vascular tumor thrombus.Low expression of miR-194-5p had a short survival time by survival analysis.Positive correlation was found between miR-194-5p and E-cadherin(r=0.68,P=0.014).Negative correlaton was found between miR-194-5p and N-cadherin(r=-0.62,P=0.013).The expression of E-cadherin was increased in miR-194-5p group than that blank control group and empty vector group.The expression of N-cadherin was decreased in miR-194-5p group than that blank control group and empty vector group.Conclusion:Lower expression of miR-194-5p was found in cholangiocarcinoma,which is related to the development and biological behavior.miR-194-5p has a targeted relationship with E-cadherin,which has a certain value in regulating cell adhere.miR-194-5p may be an independent prognostic factor for cholangiocarcinoma.
