Abstract:
Objective:To investigate the effect of STIM1 on proliferation and metastasis of osteosarcoma 143B cells and its underlying mechanism.Methods:STIM1 was silenced in 143B cell line through shRNA.The expression of STIM1 was detected by Western Blot and real-time PCR,respectively.The effect of STIM1 on the proliferation of 143B was detected by colony formation assay and CCK-8.The effect of STIM1 on the migration and invasion of osteosarcoma cells was observed by Transwell assay.The protein levels of matrix metalloproteinases MMP-2 and MMP-9 were detected by Western Blot.Results:After STIM1 was silenced in osteosarcoma cells,the proliferation capacity of the cells decreased significantly.In addition,downregulation of STIM1 effectively inhibited the migration and invasion of osteosarcoma cells,and downregulated the expression protein levels of MMP-2 and MMP-9 in 143B cells.Conclusion:Knockdown of STIM1 can effectively downregulate the protein levels of MMP-2 and MMP-9,thereby inhibiting cell proliferation,migration and invasion of osteosarcoma,which may be a new therapeutic target for osteosarcoma.
