Abstract:
Objective:To investigate the effects of miR-338-3p and PTP1B on the migration and invasion of gastric cancer cells.Methods:Western Blot was used to test the expression of PTP1B in gastric cancer specimens and adjacent tissues.qRT-PCR was used to detect the expression of miR-338-3p.SGC7901 cells were transfected by PTP1B plasmid and siRNA,miR-338-3p mimics.Transwell assay was used to detect cell migration and invasion.Finally,we verified the targeting of PTP1B by miR-338-3p by the dual luciferase reporter gene.Results:In this study,we found that PTP1B protein level was obviously increased in gastric cancer tissues and that PTP1B promoted the migration and invasion of gastric cancer cell lines.Bioinformatics analysis identified that,miR-338-3p might potentially target PTP1B.In gastric cancer tissue,miR-338-3p was negatively correlated with PTP1B (r=-0.653,P=0.013).We demonstrated that miR-338-3p regulated PTP1B in gastric cancer cells and that it regulated the migration and invasion of gastric cancer cells.Conclusion:miR-338-3p can inhibit the migration and invasion of gastric cancer cells,which may be related to targeting PTP1B,which may provide new insights into novel molecular therapeutic targets for gastric cancer.
