Abstract:
Objective:To detect the expression of calcitonin α2δ1 and miR-107 in laryngeal squamous cell carcinoma,explore their possible role in the pathogenesis of laryngeal cancer.Methods:We collected 40 cases of laryngeal cancer patients and adjacent normal tissues,and detected the expression of α2δ1 and its coding genes CACNA2D1 and miR-107 in two tissues,and analyze whether their expression is related to the clinical characteristics and prognosis of patients.Bioinformatics technology was used to find potential binding sites of CACNA2D1 and miR-107.The dual-luciferase reporter gene was used for verification.Cell migration and proliferation experiments were performed after overexpression and knockdown of miR-107 using laryngeal cancer cell TU212.Results:The results of immunofluorescence and Western blot analysis showed that the expression of α2δ1 was higher in laryngeal carcinoma than in adjacent normal tissues (P<0.05).And the RT-qPCR analysis showed that the expression level of miR-107 was significantly decreased in laryngeal carcinoma tissues compared with adjacent normal tissues (P<0.05).The low expression of miR-107 was associated with lymph node metastasis,tumor differentiation and primary site in patients with laryngeal cancer (P<0.05),but not with age and tumor TNM stage (P>0.05).The high expression of CACNA2D1 was closely related to lymph node metastasis,tumor differentiation and TNM staging in patients with laryngeal cancer (P<0.05),but not related to the age of LSCC patients and the primary site of tumor (P>0.05).The expression of CACNA2D1 was correlated with patients' relapse and three-year survival rate (P<0.05),while miR-107 was not correlated with it (P>0.05).Dual-luciferase reporter gene experiments confirmed that miR-107 can bind to two sites on the 3′-UTR end of CACNA2D1.Overexpression of miR-107 in TU212 cells inhibited cell migration and proliferation,and knockdown enhanced migration and proliferation of laryngeal cancer cells (P<0.05).Conclusion:Calcium channel protein α2δ1 plays a cancer-promoting role in patients with laryngeal squamous cell carcinoma.miR-107 can directly target CACNA2D1 to regulate the migration and proliferation of laryngeal cancer cells.
