Abstract:
Objective:To investigate the effect of SPOCK2 gene up-regulation on the proliferation of the prostate cancer cells.Methods:Real-time PCR and Western Blot was used to detect the expression of SPOCK2 in the prostate cancer tissues and cells.The expression of SPOCK2 gene in prostate cancer cell lines DU145 and LNCaP was upregulated through gene recombination.Cell proliferation was detected by CCK8 assay,and the cell cycle and apoptosis were detected by flow cytometry.Results:The expression of SPOCK2 was significantly lower in prostate cancer tissues than that in prostate hyperplasia tissues (P<0.05).The expression of SPOCK2 gene was effectively upregulated by gene recombination,and the proliferation rate of the transfected group was significantly lower than that of the negative control group and the vector control group (P<0.05).The percentage of G0/G1 phase cells in the transfection group was significantly higher than that in the negative control group and the vector control group (P<0.05).The S phase of the transfection group was significantly lower than that of the negative control group and the vector control group (P<0.05).The apoptosis rate of DU145 cells of transfected group was significantly higher than that of the negative control group and the vector control group (P<0.05).Conclusion:Up-regulation of SPOCK2 can inhibit cell proliferation,promote apoptosis,and arrest cells in G0/G1.SPOCK2 may play an important role in the development of prostate cancer by affecting cell proliferation.
