Abstract:
Objective:To explore the expression of USP9X in colon cancer tissue and the effect of USP9X on proliferation of colon cancer cell.Methods:A total of 76 colon cancer patients who underwent surgery in general hospital of central war zone PLA was included.The expression of USP9X in colon cancer tissue and adjacent normal colon tissue was tested by immunohistochemistry.Then siRNA targeting USP9X was transfected into colon cancer HCT116 cell,and the proliferation ability of HCT116 cell was tested by MTT method.Results:There were 53 patients with USP9X positive cases among 76 colon cancer patients,and the positive expression rate was 69.7%.The expression of USP9X was found to be associated with lymph node metastasis and tumor invasion.After USP9X expression of HCT116 cell was suppressed by interference siRNAs,HCT116 cell proliferation was inhibited significantly.Conclusion:It was found that positive expression rate of USP9X in colon cancer tissue was higher than that in distal normal mucosa tissue.The increased expression of USP9X was probably associated with metastasis and tumor invasion.Downregulation of USP9X decreased the proliferation ability of colon cancer cell.
