Abstract:
Objective:To observe the expression of Beclin1 and LC3B in the gastric adenocarcinomas with different differentiation degree tissues and the association of Beclin1,LC3B with Hp infection.Methods:HE and immunohistochemistry stainings were used to detect the expression of Beclin1,LC3B and Hp in the gastric adenocarcinoma tissues(87 cases)and in the chronic non-atrophic gastritis tissues surrounding the gastric adenocarcinomas(87 cases).Results:The high level expression rates of Beclin1,LC3B in the gastric adenocarcinoma group were 64.4%(56/87),62.1%(54/87)respectively,and compared with those in the chronic non-atrophic gastritis group,there were statistically significant differences respectively(P<0.001,P<0.001).Compared with those of Beclin1,LC3B in the poorly differentiated adenocarcinom group[(47.8%(11/23),30.4%(7/23)],the high level expression rates of Beclin1,LC3B in the well-moderately differentiated adenocarcinoma group[70.3%(45/64),73.4%(47/64)] were higher,and there were statistically significant differences respectively (P<0.05,P<0.05).Immunohistochemistry showed that there was significantly positive correlation between the dot shaped positive expression of LC3B and the high level expression of LC3B in the corresponding gastric adenocarcinoma(r=0.479,P<0.001).The expression of Beclin1 in the gastric adenocarcinomas was positively related with the expression of LC3B(r=0.408,P<0.001).The high level expression rates of Beclin1,LC3B in the Hp(+)gastric cancer tissues (79.59%,75.51%)were significantly higher than that in the Hp(-)gastric cancer tissues(44.74%,44.74%),and there were significant differences between them(P<0.05,P<0.05).There were significantly positive correlations between the Hp positive expression and the high level expression rates of Beclin1,LC3B in the gastric adenocarcinoma tissues(r=0.286,P<0.05;r=0.298,P<0.05).Conclusion:The increased expression of Beclin1,LC3B in the gastric adenocarcinoma tissues,especially in the well-moderately differentiated adenocarcinoma tissues,suggested that Beclin1,LC3B participated in the procession of the gastric epithelial cells' malignant transformation,progression and differentiation.The result of Hp infections significantly correlating with the high level expression rates of Beclin1,LC3B in the gastric adenocarcinoma tissues pointed out that Hp infections could induced autophagy activity increasing and they interacted with each other which could effect the development of the gastric adenocarcinoma.
