Abstract:
Objective:To investigate the expression of dipeptidase 1 (DPEP1) mRNA and protein in colorectal intraepithelial neoplasia and its relationship with clinical characteristics.Methods:Real-time fluorescence quantitative reverse transcription PCR(RFQ-RT-PCR) was used to detect the expressions of DPEP1 mRNA in a total of 60 cases of colorectal intraepithelial neoplasia (35 cases of adenomas and high-grade intraepithelial neoplasia,25 cases of adenoma and low level intraepithelial neoplasia),and 30 cases of normal colorectal mucosa epithelial tissues and 20 cases of well-differentiated adenocarcinomas,as well as their relationships with clinicopathological features were analyzed.Immunohistochemistry was used to detect the distribution and expression of DPEP1 protein.Results:The expression levels of DPEP1 mRNA in the high level traepithelial neoplasia group and low level intraepithelial neoplasia group were 0.560±0.052 and 0.328±0.040 respectively (P=0.001).The expression levels of DPEP1 mRNA in 60 cases of colorectal intraepithelial neoplasia,30 cases of normal colorectal mucosa epithelial tissues and 20 cases of well-differentiated adenocarcinomas were 0.684±0.108,0.235±0.045 and 0.832±0.136 respectively (P<0.001).The expression rate of DPEP1 in well-differentiated adenocarcinomas of was 100%(20/20),significantly higher than the 83.33%(50/60) in colorectal intraepithelial neoplasia and 20%(6/30) in normal colorectal mucosa epithelial tissue,diffusely distributing in the cytoplasm (P<0.01).The positive expression rate of DPEP1 in the high level traepithelial neoplasia group and low level intraepithelial neoplasia group were 88.57%(31/35),76%(19/25)respectively.There were correlation between DPEP1 expression level and intraepithelial neoplasia pathologic stage and advance (P<0.05),but there were no correlation with gender and age (P>0.05).Conclusion:DPEP1 is expressed in the colorectal intraepithelial neoplasia and early stage colorectal carcinoma and affects tumor cell invasiveness,associated with the advance of patients with tumor.
