Abstract:
Objective:To observe the inhibitory effect of sinomenine on colon cancer cells.Methods:HCT116 cells were divided into 3 groups,and the cells were treated with DMSO(control group),20 μmol/L and 40 μmol/L of sinomenine and 6 μmol/L 5-FU respectively.The effects of different concentrations of sinomenine on the proliferation of HCT116 cells were detected by MTT.PI staining and JC-1 staining were used to detect the effects of sinomenine on cell cycle and apoptosis respectively.The effects of sinomenine on Cyclin D1 and bcl-2 in HCT116 cells were detected by Western blot.The effect of sinomenine on the migration of HCT116 cells was detected by Transwell.The effects of sinomenine on MMP2 in HCT116 cells were detected by Western blot.Results:Compared with DMSO control group,the inhibition rates of 20 μmol/L and 40 μmol/L of sinomenine and 6 μmol/L 5-FU treated on cell 24 h were (33.16±6.01)%,(47.48±2.32)% and (62.31±3.26)%.Sinomenine inhibited the transformation of G1-S phase and promoted cell apoptosis.Western blot results showed that sinomenine inhibited the expression of Cyclin D1,bcl-2 and MMP2 in HCT116 cells.Conclusion:Sinomenine inhibits the growth and migration of HCT116 cells.
