Abstract:
Objective:To observe the effect of bufalin combined with adriamycin on the growth of bladder cancer T24 cells and explore the possible mechanism of bufalin combined with adriamycin antitumor effect.Methods:Different concentrations of bufalin and adriamycin were applied to T24 cells respectively.MTT assay was used to detect the rate of cell proliferation inhibition.Flow cytometry was used to detect the apoptosis rate.The expression of Bax and Bcl-2 T24 cells were treated with two groups of drugs,and the relative apoptosis indexes were detected.Results:The results of MTT showed that the combination of bufalin and adriamycin significantly increased the proliferation inhibition rate of T24 cells compared with adriamycin alone,the difference was statistically significant (P<0.05).Flow cytometry showed that when the combination of bufalin and adriamycin T24 cell apoptosis rate was significantly higher than the single application of two drugs,the difference was statistically significant (P<0.05).RT-PCR results showed that bufalin combination of adriamycin can significantly down-regulate the expression of apoptosis-inhibiting gene Bcl-2 and up-regulate the expression of pro-apoptotic gene Bax,which is significantly different from that of drug alone (P<0.05).Conclusion:Bufalin combined with adriamycin can enhance the sensitivity of adriamycin to the proliferation inhibition of T24 cells and induce apoptosis.The mechanism of anti-tumor activity may be related to the effects on the expression of Bax and Bcl-2.
