Abstract:
Objective:To explore the expression of the circular RNA (circRNA) hsa_circ_0001785 in breast cancer tissues and cell lines,and to study the effect of hsa_circ_0001785 knockdown on the proliferation,migration and invasion of breast cancer cells.Methods:The expression levels of hsa_circ_0001785 in breast cancer tissues and breast cancer cell lines (MDA-MB-231 and SK-BP-3) were measured by quantitative real-time PCR (qRT-PCR).To assess the effect of hsa_circ_0001785 on the viability and proliferation of breast cancer cells,CCK-8 and colony formation assays were performed in MDA-MB-231 cells.Transwell migration and wound scratch assays were carried out to evaluate the effect of hsa_circ_0001785 on the migration and invasion ability of MDA-MB-231 cells.Results:The expression levels of hsa_circ_0001785 were higher in breast cancer tissues and MDA-MB-231 and SK-BP-3 cells than in adjacent normal tissues and human mammary epithelial cells (MCF10A).Knockdown of hsa_circ_0001785 markedly repressed the proliferation,migration and invasion of MDA-MB-231 cells.Besides,upregulation of hsa_circ_0001785 promoted the proliferation,migration and invasion of MDA-MB-231 cells.Conclusion:Knockdown of hsa_circ_0001785 remarkably represses the proliferation,migration and invasion of MDA-MB-231 cells.
