Abstract:
Objective:To screen the molecular markers of hepatocellular carcinoma (HCC) with diabetes mellitus (DM),to detect its expression in liver cancer and diabetes, and to provide the molecular markers for early diagnosis of HCC with DM.Methods:The differentially expressed molecule in HCC and diabetic patients was screened by a comprehensive analysis of GEO and TCGA databases, and the expression of the molecular in normal tissue and HCC tissue was tested on tissue microarray.ELISA was used to detect the expression of the molecular in serum samples from diabetic patients.Clinical blood samples were collected to detect the expression of the molecular marker in normal people,HCC with DM or without DM.Then, the correlation between the molecular marker and blood glucose was analyzed in 15 patients with liver cancer. Results:By analyzing GEO and TCGA database,we got CXCL14 which was differentially expressed in HCC with DM.The results of tissue microarray and ELISA showed that the expression of CXCL14 was lower in HCC patients and diabetic patients, compared with normal people. The expression of CXCL14 in HCC with DM patients was significantly lower than that in HCC without DM.The expression level of CXCL14 was negatively correlated with the concentration of blood glucose in HCC patients.Conclusion:CXCL14 may be associated with the occurrence and development of hepatocellular carcinoma with diabetes mellitus, early detection of CXCL14 in serum of patients with hepatocellular carcinoma may be a predictor of diabetes mellitus.
