Abstract:
Objective:To explore the relationship between cytotoxic T-lymphocyte antigen-4 (CTLA-4) gene polymorphisms -318 T/C,CT60 G/A,+49 G/A and susceptibility to colorectal cancer,and provide theoretical basis for the early forecasting of colorectal cancer.Methods:The data of 167 cases of colorectal cancer patients were selected as experimetal group,and 245 cases of healthy people were also recruited as control group.Peripheral blood of each participant was collected and genomic DNA was extracted,and then,CTLA-4 -318 T/C,CT60 G/A,+49 G/A gene polymorphisms were detected by PCR and direct sequencing.The frequences of -318 T/C,CT60 G/A,+49 G/A genotypes and alleles were compared and the relationship between CTLA-4 -318 T/C,CT60 G/A,+49 G/A gene polymorphisms and colorectal cancer were analysed.Results:Compared with control group,inregular diet,defecation habits and basic diseases(polyp,inflammation) might be the high risk factors of colorectal cancers.Compared with patients carrying +49 AA genotype,the risk of patients carrying +49 GG genotype was OR=1.698.Meanwhile,the frequencies of CTLA-4 +49 G allele carriers were significantly higher in patients than in controls (OR=0.669,P=0.005).The distribution of CTLA-4 +49 G/A genotpes in different stage of colorectal cancer patients had strong statistical differences(P<0.05).Conclusion:CTLA-4 +49 G/A genotype had strong correlation with the occurrence of colorectal cancer.
