Abstract:
Objective:To explore the effects of EZH2 on the biological behavior of colon cancer cells,such as proliferation,apoptosis,invasion and migration.Methods:To design and synthese siRNA fragments(EZH2-siRNA) according to EZH2 gene sequence and transfect EZH2-siRNA into HCT116 cells.Using QRT-PCR to detect the mRNA expression.Using Western blot technology to detect protein expression level of EZH2.The proliferation of HCT116 cells was analyzed by MTT.The apoptosis of cells was analyzed by flow cytometry.Transwell cell invasion assay for detecting the invasion and migration abilities of HCT116 cells.Results:EZH2-siRNA down regulated mRNA expression level of EZH2 in HCT116 cells and inhibited the proliferation,invasion and migration abilities of HCT116 cells,and promoted cells apoptosis.Conclusion:EZH2-siRNA can down regulate the expression of EZH2,and effectively inhibit cell proliferation and invasion abilities of HCT116 cells.EZH2 may play an important role in the progression of colon cancer and become a new therapeutic target for colon cancer.
