Abstract:
Objective:To investigate the effect of silencing Dicer by RNAi on the biological characteristics of human breast cancer MCF-7 in vitro.Methods:Liposome mediated method was used to transfect siRNA sequence targeting Dicer as control into MCF-7 cells.Dicer expression levels after transfection were detected by using Real-time PCR and Western blot.The biological behavior cells of MCF-7 cells were evaluated by MTT and transwell assay.Results:Compared to control group,the mRNA levels of Dicer in siRNA group were significantly reduced at 24,48 and 72h after transfection(0.00152,0.00063,0.00096,respectively,P<0.01),with the effect most obvious at 48h after transfection.The protein levels in siRNA group were also significantly reduced(2.581±0.028,P<0.01).MTT assay revealed the absorbance value in siRNA group was significantly increased(P<0.01),and Transwell assay showed the number of migratory cells in siRNA group was significantly increased at 48h after transfection.Conclusion:Silencing of Dicer significantly enhanced proliferation and migration of MFC-7 cells.
