Abstract:
Objective:To study the re-expression of FHIT raise the apoptosis of colon cancer SW480 cells reacted by Mitomycin C.Methods:Eukaryotic vector expressing FHIT gene and empty vector were transfected into human colon cancer SW480 cells.Then,the FHIT proteins were detected by Western blotting.Cell growth inhibition reacted by Mitomycin C was determined by MTT.Cell apoptosis was determined by flow cytometry analysis.Results:Marked expression of FHIT protein was detected in SW480 cells after transfection with PRC/CMV vector,but it was not observed in SW480 cells after transfection with empty vector.Cell growth inhibition of the FHIT+ SW480 cell treated with Mitomycin C was more obvious than FHIT- SW480 cell.The apoptosis of the FHIT+ SW480 cell treated with Mitomycin C was more obvious than FHIT- SW480 cell.Conclusion:FHIT gene transfected by plasmid don't increase the apoptosis of cell,but,FHIT gene transfection can increase drug sensitivity of MMC.
