Abstract:
Objective:To investigate the effect and mechanism of diallyl disulfide(DADS) on invasion ability of esophageal cancer cells in vitro.Methods:MTT assay was executed to determine the inhibitory effect of DADS on ECA-109 cells,transwell chamber assay was used to determine DADS-induced effect on invasion ability of esophageal cancer cells.Western blot was used to detect DADS-induced effect on expression of MMP-2,MMP-9,TIMP-1,TIMP-2 protein in esophageal cancer cells.Results:DADS illustrate an inhibitory effect on ECA-109 cells over the concentration of 30μg/ml,DADS could significantly reduce the number of esophageal cancer cells through the invasion chamber in a dose-dependent manner.The expression levels of MMP-2,MMP-9 in esophageal cancer cell were reduced and at the same time the expression of TIMP-1,TIMP- 2 levels were significantly enhanced by DADS.Conclusion:DADS possess an ability of reducing invasion capacity of esophageal cancer cells,inhibiting the expression levels of MMP-2,MMP-9,and enhancing the levels of TIMP-1,TIMP-2.
